摘 要:
以黑果枸杞(Lycium ruthenicum)成熟叶片为外植体, 诱导愈伤组织及不定芽的分化, 获得再生植株, 建立了黑果枸杞组织培养和快速繁殖技术新体系。结果表明: 诱导叶片脱分化形成愈伤组织的适宜培养基为MS+6-BA 1.0 mg•L-1+NAA 0.2 mg•L-1, 诱导率100%; 诱导愈伤组织分化和生长的适宜培养基为MS+ZT 1.4 mg•L-1+NAA 0.02 mg•L-1, 平均丛生芽数为23.7株; 诱导生根的适宜培养基为MS+IBA 0.6 mg•L-1+NAA 0.4 mg•L-1, 生根率100%; 将生长良好的组培苗移栽并覆盖保湿膜一周, 成活率达92.5%。试验成功建立了黑果枸杞快速繁殖体系。关键词:黑果枸杞; 叶片分化; 组织培养; 植株再生

Abstract:
In the paper, we studied in vitro culture of Lycium ruthenicum by using leaf explants, including callus induction, adventitious shoot differentiation and plantlet regeneration. The results indicated that MS+6-BA 1.0 mg•L-1+NAA 0.2 mg•L-1 was the suitable medium for callus induction, with an induction rate of 100%; MS+ZT 1.4 mg•L-1+NAA 0.02 mg•L-1 was the optimizing medium for multiple shoot clumps, the multiplication coefficient could reach up to 23.7; MS+IBA 0.6 mg•L-1+NAA 0.4 mg•L-1 was the best medium for rooting, the rooting rate could reach to 100%. The strong rooting plantlets were transplanted into nutritive soil and covered the young plants with plastic farm film for a week, the survival rate were 92.5%. We have established a high-frequency regeneration system for L. ruthenicum.Key words: Lycium ruthenicum; leaf differentiation; tissue culture; plantlet regeneration
